Not sure if this is a UI setting or a default change, but clicking on "nothing" with the mouse usually deselects your target, but this no longer works.

It sounds like you have Sticky Targeting on. You can disable this by going to Interface -> Controls and unchecking the "Sticky Targeting" box off.
